摘要
应用原生质体融合技术将微生物絮凝剂产生菌B-6-1与高效石油降解菌SJ-1进行融合,从所得的67株融合菌株中筛选出菌株FB-1。确定最优发酵条件为:发酵时间44 h、接种量1.5%(体积分数)、培养基初始pH值8.0、培养基装液量30 mL/250 mL三角瓶、培养温度30℃。遗传稳定性实验表明,菌株FB-1遗传稳定,其石油类与COD去除率明显高于原始菌株B-6-1。
A flocculant-producing bacterium B-6-1 and oil-degrading bacterium SJ-1 were fused by protoplast fusion technology.FB-1 was screened from 67 protoplast fusants.The fermentation conditions of FB-1 were optimized as follows:the fermentation time was 44 h,the inoculum amount was 1.5%(volume fraction),the initial pH value was 8.0,the medium volume was 30 mL/250 mL,and the fermentation temperature was 30℃.The genetic characteristics of FB-1 was stable through the test.Its oil removal rate and COD removal rate were significantly higher than those of the original strain B-6-1.
