小柴胡汤对免疫抑制小鼠淋巴细胞共刺激分子的免疫调节作用 被引量:18

Immunoregulatory Activity of Bupleuri Decoction on the Expression of Costimulatory Molecules on the Lymphocytes in Immunosuppressed Mice

摘要 目的探讨小柴胡汤对免疫抑制小鼠外周血淋巴细胞共刺激分子的调节作用。方法每日给予正常小鼠及肌肉注射环磷酰胺诱导的免疫抑制小鼠小柴胡汤25g/kg体重,连续10d。并设免疫抑制模型及正常对照小鼠。于实验第11天,收集所有小鼠血样,分离淋巴细胞,采用流式细胞术检测实验小鼠外周血淋巴细胞CD28、CTLA-4、CD80和CD86的表达,并与对照组比较。结果与正常对照小鼠比较,免疫抑制小鼠淋巴细胞亚群发生明显变化、CD4/CD8比例明显倒置(P<0.01),淋巴细胞CD28、CTLA-4、CD80和CD86的表达明显降低(P<0.01)。与正常对照小鼠相比较,给予小柴胡汤的正常小鼠的淋巴细胞亚群及淋巴细胞共刺激分子的表达差异无统计学意义(P>0.05)。与免疫抑制小鼠相比较,给予小柴胡汤的免疫抑制小鼠CD3+细胞增加(P<0.01),CD3+CD4+细胞增加(P<0.01),CD3+CD8+细胞降低(P<0.01),CD4/CD8比例升高(P<0.01),CD28、CD86和CD80表达增加(P<0.01),而CTLA-4的表达无明显差异(P>0.05)。结论小柴胡汤可能通过调节淋巴细胞表面协同刺激分子的表达进而调节免疫抑制小鼠的免疫功能。 Objective To investigate the immunoregulatory effect of Bupleuri Decoction on the expression of costimulatory molecules in immunosuppressed mice.Methods Normal BALB/c mice and the immunosuppressed mice induced with cyclophosphamide were administrated with Bupleuri Decoction for 10 days.Normal mice and immunosuppressed mice (without the treatment) were used as control.At the 11th day, blood samples and lymphocytes were collected.CD28, CTLA-4, CD80 and CD86 expressed on peripheral blood lymphocytes were detected with flow cytometry.Results Compared with the normal control mice, lymphocyte subsets change differently, ratio of CD4 and CD8 is inversed (P0.01).Expression of CD28, CTLA-4, CD80 and CD86 was decrease (P0.01) in immunosuppressed mice.There was no difference in the lymphocyte subsets and the expression of costimulatory molecules between normal control mice and the normal mice administered with Bupleuri Decoction (P0.05).Compared with immunosuppressed mice, the expression of CD3+ and CD3+CD4+ lymphocytes was increased (P0.01), whereas the expression of CD3+ CD8+ lymphocytes was decreased (P0.01), and the ratio of CD4 to CD8 was elevated (P0.01).The expression of CD28, CD86 and CD80 was also increased (P0.01).However, the expression of CTLA-4 was not significantly difference (P0.05) in the immunosuppressed mice administered with Bupleuri Decoction.Conclusion Bupleuri Decoction can regulate the immune function of immunosuppressed mice through regulating the expression of costimulatory molecules.
