摘要
目的探讨不同补肾方药对大鼠BMSCs分泌NGFβ、BDNF、VEGF功能的影响作用。方法采用全骨髓贴壁法体外扩增培养BMSCs,流式细胞仪检测细胞表面抗原,ELISA法测定右归丸、左归丸、地黄饮子及中药联合RA诱导的大鼠BMSCs培养液中NGFβ、BDNF、VEGF的水平。结果第F3代BMSCs表面CD45-CD90+表达率为97%;与空白组比较,各药物组BMSCs培养液中NGFβ、BDNF、VEGF含量均明显增高(P<0.01或P<0.05);与RA组比较,地黄饮子组NGFβ、BDNF、VEGF含量明显增高(P<0.01),左归丸组VEGF含量明显增高(P<0.01);与右归丸组比较,地黄饮子组BDNF、VEGF含量明显增高(P<0.01),左归丸组VEGF含量明显增高(P<0.01);各中药联合RA组比单纯中药或RA单药组3种神经营养因子的含量均明显升高(P<0.01)。结论右归丸、左归丸、地黄饮子、RA均能明显促进BMSCs分泌功能,并中药与RA具有协同促进作用;地黄饮子在促进BMSCs分泌BDNF和VEGF、左归丸在促进分泌VEGF功能方面优于右归丸。
We aimed to explore the effects of different herbs on rat BMSCs secreting NGFβ,BDNF,VEGF.We isolated rat BMSCs from whole bone marrow,and then amplified the cells by adherent culture in vitro.Flow cytometry was used to detect cell surface antigens;ELISA was employed to determinate BMSCs levels of NGFβ,BDNF,and VEGF induced by You Gui Wan,Zuo Gui Wan,Di Huang Yin Zi and Chinese medicine combined with retinoic acid(RA) in culture medium.The results showed that the expression rate of CD45-CD90+ on passage 3 BMSCs surface was 97%.Compared with the control group,all medicine groups shown higher levels of NGFβ,BDNF,and VEGF(P 0.01 or P 0.05).As compared with that of the RA group,NGFβ,BDNF,and VEGF levels of Di Huang Yin Zi group were significantly higher(P 0.01),while VEGF of Zuo Gui Wan group was significantly higher(P 0.01);compared with You Gui Wan group,BDNF,VEGF levels of Di Huang Yin Zi Group were significantly higher(P 0.01),while VEGF of Zuo Gui Wan group was significantly higher(P 0.01).The content of three neurotrophic factors in Chinese medicine combined with RA was significantly higher than that in pure traditional Chinese medicine or RA single-agent group culture medium(P 0.01).We can conclude that You Gui Wan,Zuo Gui Wan,Di Huang Yin Zi and RA can significantly promote the secretory function of BMSCs,and Chinese herbs with RA have played a synergistic role;Di Huang Yin Zi is superior to You Gui Wan in promoting secretion of BDNF and VEGF,while Zuo Gui Wan is superior to You Gui Wan in promoting secretion of VEGF.
