摘要
目的建立抗人胆囊癌杂交瘤细胞系,对其抗体特异性进行研究。方法以胆囊癌细胞系SGC-996免疫Balb/c小鼠,运用淋巴细胞杂交瘤技术,获得稳定分泌抗SGC-996单克隆抗体的阳性杂交瘤细胞。采用免疫细胞化学和免疫组织化学方法,检测所获单抗对胆囊癌,部分消化道肿瘤以及乳腺癌、膀胱癌等其他肿瘤的特异性;Western blot检测抗体所特异结合抗原的性质。结果杂交瘤细胞符合融合细胞核型特点,有较强的成瘤性。抗体的Ig亚类为IgG3。相对应的抗原主要表达在细胞质和细胞膜,细胞核阴性。抗体对胆囊癌有较强的免疫反应。结论建立抗人胆囊癌杂交瘤细胞系,能稳定分泌单克隆抗体,该抗体具有较高特异性。
Out aim is aimed to prepare monoclonal antibodies(McAb) against human gallbladder carcinoma for further study of tumour,clinical diagnosis and drug-guided treatment.we immunized Balb/c mice were with SGC-996 human gallbladder carcinoma cells.and used lymphocytes hybridoma technique for McAb preparation.Then the character of McAb associated antigen was tested by the immunohistochemistry(IHC),immunocytochemistry(ICC) and Western blot ting.The cell line inoculated in abdominal cavities of Balb/c could develop to tumour on mesentery.and the McAb was found to be the IgG3 isotype.The antigen recognized by McAb was located mainly in cytoplasm.The ICC results showed that the McAb reacted strongly to gallbladder carcinoma but demonstrated negative reaction with non-digestive tract tumor.This research has established the hybridoma cell line which can stably secret monoclonal antibody against human gallbladder carcinoma.
