摘要
目的 P0蛋白与内耳和周围神经自身免疫性疾病的发生有关,本研究拟通过比较耳蜗神经和坐骨神经中该蛋白的一致性和mRNA水平,为自身免疫性内耳病的诊治提供线索。方法分别提取36只SD大鼠的耳蜗神经和坐骨神经总RNA,采用反转录-聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)及半定量RT-PCR测试P0蛋白的mRNA表达水平,将RT-PCR产物进行cDNA序列测定,比较大鼠耳蜗神经和周围神经P0蛋白的cDNA序列和mRNA转录水平。结果大鼠耳蜗神经和坐骨神经P0蛋白的cDNA序列相同,但二者之间的mRNA转录水平有显著性差异。结论大鼠耳蜗神经和坐骨神经的P0蛋白具有一致性,其mRNA转录水平有显著性差异。
Objective To compare myelin protein zero cDNA and mRNA levels in the cochlear and sciatic nerves in rats for possible new approaches to autoimmune inner ear diseases. Methods Total RNA was extracted from 36 rats. Expression of myelin protein zero mRNA was tested by semiquantitative RT-PCR and cDNA sequences were determined. cDNA sequences and mRNA transcription levels in cochlear and peripheral nerves were compared. Results cDNA sequences were the same in cochlear and sciatic nerves, but the mRNA levels were different. Conclusion Myelin protein zero is identical in the cochlear and sciatic nerves, although its mRNA transcription appears to be different in the two nerves.
