TLR4介导口腔鳞癌细胞分泌IL-6、IL-8及VEGF 被引量:6

Secretion of IL-6,Il-8 and VEGF in human squamous carcinoma cells induced by TLR4 ligation with LPS

摘要 采用RT-PCR和流式细胞术检测人永生化口腔上皮细胞(HIOEC)及人口腔鳞癌细胞CAL-27的TLR4的表达,观察脂多糖(LPS)刺激CAL-27细胞后TLR4表达的变化,同时使用ELISA检测细胞培养上清IL-6、IL-8、VEGF和TGF-β的变化。结果发现TLR4在人口腔鳞癌细胞CAL-27有较高的表达,而在HIOEC中表达较低;LPS刺激能够影响TLR4的蛋白表达水平,促进CAL-27细胞分泌IL-6、IL-8和VEGF,而TGF-β未受影响。以上结果提示,TLR4可能在人口腔鳞癌的发生和转移中发挥作用。 The expression of TLR4 in human oral squamous carcinoma cells CAL-27 was detected by RT-PCR and flow cytometry.Then,the change of TLR4 expression after stimulation of these cells with LPS was observed and the changes in the secretion of various cytokines,such as IL-6,IL-8,VEGF and TGF-β were verified by ELISA assay.To determine the expression of TLR4 in human oral squamous carcinoma cells and to explore its biological role,the expression of TLR4 was evaluated by reverse transcriptional polymerase chain reaction(RT-PCR) and analyzed by flow cytometry and the expression of TLR4 induced by LPS-ligand of TLR4 was verified by Western blot assay.It was found that TLR4 was expressed in the CAL-27 cells and it could be up-regulated by LPS ligation.Also,LPS promoted the production of IL-6,IL-8 and VEGF in human oral squamous carcinoma cells.Our study demonstrates that TLR4 is expressed in human oral squamous carcinoma cells and TLR4 and its ligand may play an important role in the occurrence and metastasis of human oral squamous cell carcinoma.
