喉癌组织中HIF-1α和COX-2及VEGF与微血管密度的关系及临床意义 被引量:10

Clinical significance and relationship between HIF-1α,COX-2,VEGF and microvessel density expression in laryngeal carcinoma

摘要 目的:探讨喉癌患者中缺氧诱导因子-1α(HIF-1α)、环氧合酶-2(COX-2)与血管内皮生长因子(VEGF)表达水平与喉癌血管生成的关系及临床意义。方法:通过免疫组织化学SP法对64例喉癌标本中HIF-1α、COX-2与VEGF及微血管密度(MVD)进行了检测。结果:喉癌中HIF-1α、COX-2蛋白及VEGF的阳性表达率分别为65.63%(42/64)、68.75%(44/64)和71.88(46/64),MVD为35.79±9.49。喉癌HIF-1α、COX-2和VEGF与MVD表达均呈正相关(P<0.05)。喉癌中HIF-1α和COX-2与VEGF之间存在正相关性(P<0.05)。在有淋巴结转移的肿瘤中HIF-1α、COX-2和VEGF阳性表达及MVD明显高于非转移组(P<0.05)。结论:HIF-1α、COX-2和VEGF参与喉癌的血管生成,HIF-1α通过调节下游基因的表达(COX-2及VEGF),在喉癌血管生成中发挥重要作用,共同参与喉癌颈淋巴结转移过程。 Objective:To evaluate the role of HIF-1α, C OX-2 ,VEGF proteins and Microvessel density in angiogenesis and matastasis of laryngeal carcinoma.Method:Surgically resected specimens of 64 laryngeal carcinomas were studied by immunohistochemical stainning for HIF-1α protein, COX-2 protein, VEGF and MVD.Result:Positive ex pressions of HIF-1α,COX-2 protein and VEGF were found in 65.63%, 68.75% and 71.88% of laryngeal carcinoma specimens, respectively.The mean MVD in this ser ies was 35.79±9.49.The expression of HIF-1α, COX-2 and VEGF positively cor related with MVD expression.There was a positive relationship between HIF-1α, COX-2 and VEGF expression.The microvessel density(MVD) in HIF-1α,COX-2 and VEGF positive tumors was significantly higher in patients with cervical lymph no de metastases than that in patients without metastases.Conclusion:HIF-1α,COX-2 and VEGF proteins may play an important role in the angi ogenesis of laryngeal carcinoma.HIF-1α regulate the expression and function of its downstream target genes(COX-2 and VEGF),which play a critical role in th e angiogenesis of laryngeal carcinoma, metastases in the process of laryngeal cancer cell spreading to cervical lymph nodes.
