雷公藤甲素洗脱支架对猪再狭窄模型冠状动脉增殖和内皮素的影响

摘要 目的 研究雷公藤甲素洗脱支架抑制血管新生内膜增殖的有效性、对内皮功能的影响及支架安全性. 方法 将16只冠状动脉再狭窄模型小型猪完全随机分为2组,每组8只,分别植入雷公藤甲素洗脱支架和雷帕霉素支架,12周时比较2组形态测量学、组织病理学、血浆内皮素水平及生化检测结果. 结果 雷公藤甲素组的新生内膜面积[(0.74±0.05)mm^2]与雷帕霉素组[(0.70±0.04)mm^2]相似,但差异无统计学意义(P>0.05).雷公藤甲素组4周的内皮素血浆水平[(76.1±20.5)ng/L]低于雷帕霉素组[(88.3±17.9)ng/L],但差异无统计学意义(P>0.05).血生化检查未发现雷公藤甲素洗脱支架对受试动物产生毒副作用. 结论 雷公藤甲素洗脱支架在12周内能够抑制新生内膜增殖,对内皮有一定保护作用,有较好的安全性. Objective To investigate the anti-proliferation and endothelial protecting effect of triptolide-eluting stent. Methods Experimental porcines were divided into 2 groups with 8 porcines in each group:triptolide-eluting stent group and rapamycine-eluting stent group.Morphometric,histopathologic,RIA and biochemical staining analysis were compared between the two groups 12 weeks after stenting in porcine model of coronary restenosis. Results The neointimal areas of triptolide-eluting stents(0.74±0.05)mm^2 were similar to rapamycine-eluting stents(0.70±0.04)mm^2(P 〉0.05).At 4 weeks,plasma endothelin-1 was lower in triptolide group(76.1±20.5)ng/L than that in rapamycine group(88.3±17.9)ng/L.No evidence of toxic effects had been found in the experimental animals related to triptolide-eluting stent. Conclusions Triptolide-eluting stent may inhibit neointimal prolieration during 12 weeks.Triptolide probably takes part in protecting endothelial function.
