摘要
目的 确定关节洗药内主药当归中阿魏酸的高效液相色谱法(HPLC)含量测定的最佳条件,并以阿魏酸为参考物,研究关节洗药的透皮吸收情况. 方法 以当归中的阿魏酸的含量为检测标准,以大鼠腹部皮肤为受试皮,采用单隔室静态小室法,测量关节洗药的透皮吸收率.HPLC对当归中阿魏酸定量测定的最佳条件为流动相:甲醇:乙腈:1%乙酸(20:10:70),检测波长320 nm,流速1.0 ml/min. 结果 阿魏酸的标准曲线为A=0.88×10^-2C-4.62×10^-2,r=0.9999,低中高三个浓度的日内精密度相对标准偏差 (RSD)为2.53%,2.62%,2.47%(n=5),日间精密度RSD为3.43%,2.88%,1.88%(n=5).加样回收率(97.00±1.92)%,RSD为1.97%.关节洗药溶液含阿魏酸(9.79±0.02)mg(n=4),RSD为0.20%.关节洗药24 h的透皮吸收率为47.12%. 结论 关节洗药中主要成分透皮吸收良好,如果能改变剂型,延长药物透皮时间,将取得更好疗效.
Objective To study the transdermal absorption of Guanjie Xiyao through rat skin with ferulic acid as the reference material. Methods The contents of HPLC of ferulic acid to rat abdominal skin with TK-12B-type diffusion cell were determined. Results Using this HPLC system,we found the Standard curve was A=0.88 × 10-2C -4.62 ×10-2,r=0.9999,and the average recovery was 97.00%,with RSD of 1.97%.Through the calculation of HPLC data,the contents of Felic acid in Guanjie Xiyao was 9.79 mg(n =4),with RSD of 0.20%.The transversal absorption rate of Guanjie Xiyao within 24 h was 47.12%. Conclusion These results demonstrate that major components of Guanjie Xiyao transdersal absorption were effedtive.Changing the dosage forms and prolonging the time of transdermal can get higher efficacy.
