摘要 目的探讨X-连锁凋亡抑制蛋白(XIAP)表达在鼻咽癌发生发展过程中的作用,以及XIAP表达与鼻咽癌分化程度和淋巴转移的关系。方法应用RT-PCR法检测45例鼻咽癌组织、17例正常鼻咽组织和鼻咽癌细胞株5-8F、CNE-2中XIAP mRNA的表达,并通过免疫组化SP法检测鼻咽癌组织及癌旁组织XIAP蛋白的表达情况。结果 XIAP mRNA在5-8F、CNE-2和鼻咽癌组织中呈高表达,正常鼻咽组织不表达或低表达;鼻咽癌组织和正常鼻咽组织XIAP mRNA阳性检出率分别为86.67%(39/45)和35.29%(6/17)(P<0.01);在鼻咽癌组织和癌旁组织中,XIAP蛋白阳性检出率分别为82.22%(37/45)和24.44%(11/45)(P<0.01);鼻咽癌组织XIAP蛋白表达与肿瘤分级和淋巴转移无关(P>0.05)。结论 XIAP在鼻咽癌中高表达,可能在鼻咽癌发生过程中具有重要的作用。 ObjectiveTo study the role of the XIAP expression in nasopharyngeal carcinoma development and the relationship between its expression and tumor grade and lymphatic metastasis.MethodsThrough RT-PCR measures,XIAP mRNA expression was detected in nasopharyngeal carcinoma tissues of 45 cases,normal nasopharyngeal tissues of 17 cases and two nasopharyngeal carcinoma cell lines(5-8F,CNE-2).Immunohistochemical SP method was used to examine the expression of XIAP protein in and around the nasopharyngeal carcinoma tissues.ResultsXIAP mRNA was highly expressed in nasopharyngeal carcinoma tissues and cell lines 5-8F,CNE-2 but not expressed or lowly expressed in normal nasopharyngeal tissues.The XIAP mRNA positive rates of nasopharyngeal carcinoma and normal nasopharyngeal tissues were 86.67%(39/45) and 35.29%(6/17),respectively(P〈0.05).XIAP protein positive rates of nasopharyngeal carcinoma tissues and adjacent tissues of nasopharyngeal carcinoma were 82.22%(37/45) and 24.44%(11/45),respectively(P〈0.05).The expression of XIAP protein in the tissue of nasopharyngeal carcinoma was not correlated with tumor grade or lymphatic metastasis(P〈0.05).ConclusionThe high expression of XIAP in nasopharyngeal carcinoma may play an important role in the development of nasopharyngeal carcinoma.
