摘要
采用苯酚-硫酸法测定山茱萸中多糖的含量,正交实验优化显色条件,确定5%苯酚用量1.2mL,浓硫酸用量7.0mL,反应时间20min。标准曲线回归方程为A=6.0393C-0.0148,r=0.9994(n=6),在(10~100)μg/mL范围内呈良好线性关系。平均回收率为104.93%,RSD=1.87%(n=6)。测得多糖平均含量为45.2%,RSD=1.72%(n=6)。该法快速、准确、灵敏,可为山茱萸中多糖的测定、开发利用和质量评估提供参考。
The polysaccharides contents in Comus officinalis Sieb.et Zucc.were determinated by phenol-sulfuric acid method.The main factors affecting the method were optimized and the results showed that the optimal conditions were the time of reaction of 20 min and the dosage of 5% phenol and H2SO4 of 1.2 and 7.0 mL,respectively.Linear range was(10~100) μg/mL(A=6.039 3 C-0.014 8,r=0.999 4,n=6).The average recovery was 104.93%,RSD was 1.87%(n=6).The polysaccharide content was 45.2% and RSD was 1.72%(n=6).It is conclused this method is rapid,reliable and sensitive.
