摘要
目的探讨苏木中药对移植心脏的保护作用及可能的作用机制。方法建立大鼠腹腔同种异位心脏移植模型。实验分为3组:A组为空白对照组;B组为苏木(Caesalpinia sappan,CS)中药组;C组为环孢素(CsA)组。每组8只大鼠。术后第2天开始灌药,第7天取供心。Western blot法检测心肌组织中NF-κB及IκBα的表达水平。结果与移植对照组相比CsA及CS组移植物的病理损害明显减轻;NF-κB及IκBα的表达水平给药组较对照组差异显著(P<0.05),但两种药物组之间的差异不显著(P>0.05)。结论苏木中药可能通过抑制NF-κB信号传导途径的激活,减缓急性排斥反应的发生从而减轻移植物的病理损伤,发挥保护移植心脏的作用。
Objective To discuss the protective effect and possible mechanism of Caesalpinia sappan on the transplanted heart.Methods The model of cardiac allotransplantation was established.The cardic allograft rats were randomly divided into 3 groups: control group(A group),Caesalpinia sappan group(B group),and cyclosporin A group(C group).Animals were given drugs 2 days after operation.The expression of NF-κB P65 and IκBα were measured 7 days after transplantation by Western blot.Results The allogeneic graft survival time were significantly prolonged in CsA and CS groups.The level of NF-κB P65 and IκBα in CsA and CS group had a significant difference compared with control group(P0.05),but the difference between the two medicines was not significant(P0.05).Conclusion Caesalpinia sappan may inhibit the activation of NF-κB signal transduction pathway,thereby slow down the acute rejection and reduce the pathological injury of transplanted heart.
