摘要
目的研究三氧化二砷(As2O3)对裸鼠肝癌侵袭及转移基因的影响。方法建立人肝癌细胞系(Bel-7402)裸鼠肝癌模型,用不同剂量As2O3治疗,计算各组瘤体体积及瘤重,计算瘤重抑制率;肿瘤组织及双肺病理组织检查,观察肿瘤血管分布,肺转移结节计数;采用免疫组化检测肿瘤组织中VEGF、COX-2、HIF-1α和CD44V6的表达情况。结果 As2O3治疗组平均瘤重和瘤体积较对照组明显减低,有显著性差异(P<0.01);病理结果,可见肝脏部位白色肿瘤组织块,并可见肿瘤表面红色粗大的肿瘤血管。As2O3治疗组与对照组相比,肺转移结节数目明显减少,有显著性差异(P<0.01)。免疫组化结果,各组细胞均着色,治疗组VEGF、COX-2、HIF-1α和CD44V6阳性表达率较对照组均降低,有统计学意义(P<0.05)。结论 As2O3能抑制肝癌细胞的侵袭和转移,降低VEGF、COX-2、HIF-1α和CD44V6的表达。
Objective To study the effects of arsenic trioxide(As2O3) on invasion and metastasis of tumor model in nude mice.Methods Hepatoma cell line(Bel-7402 cells)was established.The size and weight of tumor,inhibition ratio of tumor weight were calculated.The pathology of tumor tissues and lung tissues were examined.Pulmonary metastasis tubercle were counted.The expression of VEGF,COX-2,HIF-1α,and CD44V6 were detected by immunohistochemistry.Results The size and weight of tumor were significantly decreased in As2O3 group compared with those in control group(P0.01).White tumor tissue and red fat tumor vessel were found by pathology.The number of pulmonary metastasis tubercle after As2O3 effect was obviously lower than that of control group(P0.01).The As2O3 groups had low expression of VEGF,COX-2,HIF-1α,and CD44V6(P0.05).Conclusion As2O3 can inhibit invasion and metastasis of hepatoma,further inhibit the expression of VEGF,COX-2,HIF-1α,and CD44V6.
