乌司他丁对非酒精性脂肪肝大鼠肝脏保护作用的研究

摘要 目的研究乌司他丁对非酒精性脂肪肝(nonalcoholic fatty liver,NAFL)大鼠肝脏的保护作用。方法取24只Wistar大鼠随机分为正常对照组,高脂饮食生理盐水处理组(模型组)及乌司他丁治疗组(乌司他丁组),各8只。正常对照组和模型组大鼠每天腹腔注射生理盐水2 mL,乌司他丁组大鼠腹腔内注射含乌司他丁(4×104IU/kg)的生理盐水2 mL,于第8周处死大鼠,取肝组织用于病理组织学观察及脂肪沉积评价;取血备于血清甘油三酯(TG)、胆固醇(TC)、天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)等生化指标检测。结果模型组大鼠血清TG、TC、ALT及AST含量显著高于对照组;病理组织学证实非酒精性脂肪肝模型建立成功。乌司他丁治疗组大鼠血清TG、TC含量显著低于模型组;治疗组大鼠血清ALT及AST水平介于对照组与模型组之间,ALT值显著低于模型组,P<0.05;AST值治疗组与模型组之间无明显差别,P>0.05;光镜下观察乌司他丁治疗组肝细胞脂肪变较模型组轻。结论乌司他丁可以控制高脂饲养大鼠血脂升高,保护肝脏功能,从而抑制NAFL的形成。 Objective To study the protective effect of ulinastatin in nonalcoholic fatty liver disease(NAFLD).Methods Twenty-four Wistar rats were randomly divided into 3 groups,including model group,treat group and normal control group.Two milliliter physiological saline was injected into the abdominal cavity of normal control group rat and model group rat,and two milliliter physiological saline containing ulinastatin was injected into the abdominal cavity of treat group.These rats were executed at the 8th week,and hepatic tissue of the rats was obtained to observe the change of pathologic histology and to evaluate the degree of the fatty deposition.The blood of rats was obtained to detect some biochemical indicators such as the triglyeride(TG),cholesterol(TC),aspartate aminotransferase(AST) and alanine transaminase(ALT)in serum and so on.Results The levels of TG,TC,ALT and AST in serum were significantly higher in model group than those in control group,and the model of NAFLD was set up successfully by the confirm of pathohistology.The levels of TG and TC in serum in treat group were significantly lower than those in model group,and the levels of ALT and AST in serum in treat group were between these in the control group and the model group,but the level of ALT in serum in treat group was significantly lower than model group(P0.05).There was no significant difference in the level of ASTbetween the treat group and model group(P0.05).The result of morphology showed that the degree of fatty degeneration of liver cell was more serious in model group than that in the treat group.Conclusion UTI can control the increase of blood fat of rat raised by high fat food,protect the liver function and hold back the formation of NAFLD.
