摘要
目的探索和建立永生化的人皮肤成纤维细胞,为皮肤成纤维细胞在临床医学上的应用提供可靠的科学依据。方法胰蛋白酶消化法分离培养人皮肤成纤维细胞;细胞生长曲线及MTT法测定细胞增殖能力;流式细胞仪测定成纤维细胞生长周期及细胞增殖指数。结果不同代次原代及冻存复苏后的人皮肤成纤维细胞增殖能力均很强。结论人皮肤成纤维细胞在增殖能力上完全可以作为体细胞基因治疗的靶细胞,而且是未来最有希望的永生化基因治疗细胞之一。
【Objective】 To explore and establish immortalized huamn skin fibroblasts and to provide a reliable scientific basis for the skin fibroblasts.【Methods】Huamn skin fibroblasts were cultured by trypsin digestion.Pro-liferation were observed by growth curve and MTT.Cell cycle were observed by flow cytometer analysis and cell pro-liferation index.【Results】Huamn skin fibroblasts had better proliferate whether different passages or after cryop-reservation.【Conclusion】Therefore human skin fibroblast cells in the proliferative capacity can be used as target cells of gene therapy and the most promising cell of gene therapy.
