Ezrin和Fascin蛋白在血管瘤中异常表达的研究 被引量:1

ABNORMAL EXPRESSION AND CLINICAL SIGNIFICANCE OF EZRIN AND FASCIN IN HUMAN DERMAL HEMANGIOMA

摘要 目的应用免疫组织化学方法检测不同时期血管瘤组织中膜细胞骨架链接蛋白Ezrin和Fascin蛋白的表达,以探讨其在血管瘤发生、发展过程中的作用机制。方法收集武汉大学人民医院病理科2005年9月-2009年12月皮肤毛细血管瘤存档蜡块50例,其中男性25例,女性25例。采用免疫组织化学S-P法检测50例皮肤血管瘤增生期、退化期及正常皮肤组织中Ezrin和Fascin的表达水平,采用HPIAS-1000图文报告管理系统对Ezrin和Fascin的表达进行定量分析,并用SPSS11.5软件对各组免疫组织化学反应阳性颗粒的平均光密度、阳性面积率做单因素方差分析和SNK(q)检验。结果 (1)Ezrin的表达增生期血管瘤血管内皮细胞中可见密集分布的棕黄色颗粒,Ezrin呈高表达,正常皮肤组及退化组血管内皮细胞中可见少量的棕黄色颗粒,Ezrin表达弱。增生期组Ezrin的表达明显高于退化期组和正常皮肤组(P<0.05),而后两组比较差异无统计学意义(P>0.05)。(2)Fascin的表达增生期血管瘤血管内皮细胞中可见密集分布的棕黄色颗粒,Fascin呈高表达,正常皮肤组及退化组血管内皮细胞细胞中可见少量的棕黄色颗粒,Fascin表达弱。增生期组Fascin的表达明显高于退化期组和正常皮肤组(P<0.05),而后两组比较差异无统计学意义(P>0.05)。结论Ezrin和Fascin在血管瘤增生期均上调表达,表明Ezrin和Fascin均与血管瘤的发生、发展有密切关系。 Objective To investigate the role of Ezrin and Fascin in the development fo human dermal hemangioma.Methods Specimens from 50 cases(25 male and 25 female) of hemangioma from the Department of Pathology,Renmin Hospital,Wuhan University,from 2005 to 2009 were collected.We examined the expressions of Ezrin and Fascin in proliferative,involuting hemangiomas and normal skin tissues by using immunohistochemistry.The average optical density and the positive area rate of Ezrin and Fascin expression were measured by image analysis(HPIAS-1000).Results 1.Ezrin showed remarkably higher expression in the proliferative phase than in the involuting phase and in normal skin tissues(P〈0.05).No significant difference was found between the expression of Ezrin in involuting hemangioma and that in normal skin tissues(P〉0.05).2.Fascin also showed remarkably higher expression in the proliferative phase than in the involuting phase and in normal skin tissues(P〈0.05).No significant difference was found between the expression of Fascin in involuting hemangioma and that in normal skin tissues(P〉0.05).Conclusion Ezrin and Fascin expressions are up-regulated in proliferating hemangioma,indicating that both Ezrin and Fascin are closely related with the occurrence and development of hemangioma.
