Fractalkine对类风湿关节炎成纤维样滑膜细胞增殖的影响

The role of fractalkine in the proliferation of fibroblast-like synoviocytes obtained from rheumatoid arthritis

目的明确fractalkine(CX3CL1)对人类风湿关节炎成纤维样滑膜细胞(RA-FLS)增殖的影响。方法通过组织块法培养RA-FLS。分别用0、50、100ng/mL重组人fractalkine刺激RA-FLS24h,用四唑盐(MTT)法检测其吸光度值,比较各组间增殖率。结果 50ng/ml及100ng/ml fractalkine均可促进RA-FLS增殖(P=0.005,P=0.022)。结论 Fractalkine可促进RA-FLS的增殖,可部分解释RA-FLS自主增殖的特性。

Objective To observe the effect of fractalkine（CX3CL1） on the proliferation of fibroblast-like synoviocytes（FLS） obtained from rheumatoid arthritis（RA）.Methods RA-FLS were gained through tissue culture.Use fractalkine to stimulate RA-FLS in concentration of 0,50,100 ng/mL for 24 h,test the absorbance by the MTT cell proliferation assay,and compare the growth rate among each group.Result Fractalkine stimulates cell growth in RA-FLS at concentration of 50 or 100 ng/mL（P=0.005,P=0.022）.Conclusion Fractalkine can promote the proliferation of RA-FLS,and it partly explains the characteristic of self duplication in RA-FLS.
