姜黄素通过抑制NF-κB信号通路增敏阿霉素的抗肝癌作用 被引量:5

Curcumin Could Potentiate the Treatment Effect of Doxorubicin on Human Hepatocellular Carcinoma by Inhibiting NF-κB Signaling Pathway

摘要 目的探讨姜黄素增敏阿霉素治疗肝癌的效果和机制。方法 MTT法检测联合用药前后姜黄素和阿霉素对HepG2细胞抑制率的影响。应用Western-blot法检测联合用药前后NF-κB信号通路及其下游蛋白Bcl-2的表达,以及与内源性凋亡相关的Caspase的表达。结果联合用药后药物对HepG2细胞的抑制率明显增高,并且NF-κB信号通路的核心蛋白p65以及Bcl-2都发生明显下调。此外,与内源性凋亡相关的Caspase得到激活凋亡效果增强。结论姜黄素在体外与阿霉素联合应用,可以降低NF-κB信号通路的表达,增敏阿霉素的抗肝癌作用。 Objective To investigate the combine treatment effect of curcumin and doxorubicin on human hepatocellular carcinoma cell line HepG2.Methods Cell proliferation assay and western-blotting were analyzed in vitro.Results The anti-tumor effect of doxorubicin was enhanced by curcumin and constitutive activation of NF-κB and Bcl-2 was inhibited by the combine treatment.Furthermore,activation of caspase 3,caspase 9 and PARP was also found.Conclusion Curcumin could potentiate the anti-tumor effect of doxorubicin through inhibiting the NF-κB pathway.
