摘要
目的:探讨TKTL1在食管鳞状细胞癌中的表达及其临床意义.方法:分别采用原位杂交及免疫组织化学的方法检测62例食管鳞癌组织、31例癌旁不典型增生组织及62例正常食管黏膜组织中TKTL1蛋白及其mRNA的表达.结果:正常黏膜组织、癌旁不典型增生组织及食管鳞癌组织中TKTL1蛋白阳性表达率依次增高,分别为17.7%(11/62)、29.0(9/31)、53.2%(33/62),组间比较差异有统计学意义(均P<0.05);TKTL1蛋白表达与癌的浸润深度、淋巴结转移及TNM分期密切相关,组间比较差异有统计学意义(均P<0.05).正常黏膜组织、癌旁不典型增生组织及食管鳞癌组织中TKTL1mRNA的阳性表达率依次增高,分别为4.8%(3/62)、22.6%(7/31)、43.5%(27/62),组间比较差异有统计学意义(均P<0.05);TKTL1mRNA表达与癌的浸润深度、淋巴结转移及TNM分期密切相关,组间比较差异有统计学意义(均P<0.05).TKTL1蛋白及其mRNA的表达呈正相关关系(γp=0.302,P<0.01).结论:TKTL1蛋白及其mRNA表达率升高可能与食管鳞癌的发生、发展及浸润、转移有关.
AIM: To investigate the expression of transketolase like-1 (TKTL1) protein and mRNA and to analyze its clinical significance in esophageal squamous cell carcinoma (ESCC). METHODS: The expression of TKTL1 protein and mRNA was detected by immunohistochemistry and in situ hybridization in 62 ESCC specimens, 31 tumor-adjacent atypical hyperplastic epithelial specimens, and 62 normal esophageal mucosal specimens, respectively.RESULTS: The positive rates of TKTL1 protein expression in normal esophageal epithelium, tumor-adjacent atypical hyperplastic epithelium and ESCC were 17.7% (11/62), 29.0% (9/31) and 53.2% (33/62), respectively, with significant differences among the three groups (all P 0.05). TKTL1 protein expression was closely correlated with infiltrative depth, lymphatic metastasis and TNM grade in ESCC (all P 0.05). The positive rates of TKTL1 mRNA expression in normal esophageal epithelium, tumor-adjacent atypical hyperplastic epithelium and ESCC were 4.8% (3/62), 22.6% (7/31) and 43.5% (27/62), respectively, with significant difference among the three groups (all P 0.05). TKTL1 mRNA expression was closely associated with infiltrative depth, lymphatic metastasis and TNM grade in ESCC (all P 0.05). There is a positive correlation between TKTL1 mRNA and protein expression (γp = 0.302, P 0.01). CONCLUSION: TKTL1 expression may be associated with the development, invasion and metastasis of ESSC.
