摘要
目的通过细胞培养,观察洛伐他汀(lovastatin,LOV)对小鼠巨噬细胞株RAW264.7的作用,探讨LOV对小鼠巨噬细胞株(RAW264.7)增殖和凋亡的影响。方法以不同浓度(20,40,80μmol/L)LOV处理RAW264.7,作用不同时间(24 h,48 h,72 h),以MTT法检测LOV对小鼠巨噬细胞RAW264.7增殖的影响;观察洛伐他汀对小鼠巨噬细胞RAW264.7凋亡的影响。结果洛伐他汀对小鼠巨噬细胞瘤RAW264.7有明显的抑制生长的作用(P<0.05),细胞凋亡形态观察显示,经80μmol/L洛伐他汀作用的细胞出现明显固缩的凋亡小体。结论洛伐他汀能有效抑制巨噬细胞株的体外生长,其作用与剂量、时间具有一定的依赖性,这种抑制作用可能与洛伐他汀诱导细胞凋亡有关。
Objective: To explore the effect of lovastatin(LOV) on proliferation and apoptosis of RAW264.7.Methods: After the treatment of RAW264.7 with various concentration LOV(20μmol/L,40μmol/L,80μmol/L)for 24,48,and 72 hours,the proliferation was oberseved by MTT,the effect of LOV on apoptosis of RAW264.7 was observed from fluorescence microscopy.Results: LOV could inhibit the proliferation of cells effectively(P〈0.05) and apoptotic body could be observed from fluorescence microscopy.Conclusion: LOV can inhibit the proliferation of cancer cells effectively.All the effects above are dependent on dose and time.The inhibition effect may be related to LOV in inducing the cell apoptosis
