熵聚堆法拟方对非酒精性脂肪性肝炎疗效及作用机理的实验报告 被引量:1

摘要 目的:验证熵聚堆法所拟方剂对非酒精性脂肪性肝炎(NASH)的疗效及作用机理,以明确其可行性。方法:对近10年文献中治疗NASH的方剂进行熵聚堆方法处理,确立熵聚堆系列方;将SD大鼠分为正常组、熵聚堆1号方、2号方、1+2号方组、模型组、易善复组,用高脂饲料进行NASH造模,并用熵聚堆系列方及易善复治疗10周,最后检测各组肝组织脂质代谢相关因子、线粒体DNA修复酶,以及肝功能、血脂、病理等指标,并进行统计。结果:熵聚堆系列方组PPARα、PGC-1α含量均高于模型组(P<0.01),SREBP-1c、SCAP含量均低于模型组(P<0.01),熵聚堆系列方组线粒体DNA修复酶含量高于正常组(P<0.01),熵聚堆系列方可以降低肝脏酶谱,降低血脂,减少肝脂比(P<0.01),改善肝脏病理损害。结论:熵聚堆系列方对NASH具有良好的治疗效果,可以干预脂质代谢相关因子从而调节脂质代谢,提高线粒体DNA修复酶促进受损肝细胞自身修复,减轻脂肪在肝脏的沉积,因此,熵聚堆法创立方剂具有可行性。 This study aimed to determine the therapeutic effect and mechanism of TCM prescriptions made by entropy partition method for NASH, as well as its feasibility. The TCM prescriptions concerning NASH were collected by the entropy partition method from the papers published over the past 10 years,and the entropy partition serial prescriptions were consequently created. SD rats were divided into 6 groups :the normal group,the No.1 entropy partition prescription group,the No.2 entropy partition prescription group,the No.l+No.2 entropy partition prescription group,the model group and the Essentiale group. The NASH models were established by high-fat diets and treated by entropy partition serial prescriptions and Essentiale for ten weeks. Such factors as lipid metabolism, mitochondrion DNA repair enzymes in liver were tested, and liver functions, blood lipids, and pathological indicators were measured. All data were analyzed. PPARα and PGC-1α in the entropy partition serial prescriptions groups were higher than those in the model group(all P〈0.01 ). Mitochondrion DNA repair enzymes in the entropy partition serial prescriptions groups were higher than those in the normal group (all P〈0.01 ).Entropy partition serial prescriptions reduced hepatitis Liver enzymes, blood lipids, and the ratio of fat in liver (all P〈0.01 ), and also mitigated the pathological damages. The results show that entropy partition serial prescriptions have perfect therapeutic effects on NASH. They can adjust factors of lipid metabolism, facilitate self-repair of liver cells by increasing mitochondrion DNA repair enzymes, and thus decrease fat deposition in liver. Thus it is feasible to make TCM prescriptionsby the entropy partition method.
