摘要
目的 评估 D N A 探针鉴定伴放线放线杆菌( Aa) 的价值,探讨 Aa 与牙周炎临床表现间的相关性。方法 32 例成人牙周炎患者59 个部位龈下菌斑经 Aa 选择性培养基培养后,菌落分别用 D N A 探针法及生化法鉴定,并根据探针法的检出率探讨 Aa 感染与牙周炎临床参数间的关系。结果 D N A 探针法与培养法有一致性( P<0 .01 ,r = 0 .78 ,但探针法的检出率(26/59) 明显高于培养法(19/59)( P< 0 .05) ; Aa 的检出率与患者的年龄、性别无关,而与牙周组织病损程度相关。结论 D N A 探针法优于细胞生化鉴定法, Aa
Objective This study evaluated the feasibility of using a colony lift method and DNA probes to enumerate Actinobacillus actinomycetemcomitans cultured on primary isolation plates.Methods 59 subgingival plaque samples from 32 cases with adult periodontitis were detected by both Aa DNA probe and culture plus biochemical tests.The feasibility of Aa whole genomic DNA probe in clinical examination of periodontitis was evaluated,and the Aa positive rate by DNA probe assay was analyzed.Results DNA probe assay was superior to the culture method(26/59 vs 19/59,P< 0.05 ).Incidence of positive Aa was positively associated with clinical parameters,especially closely with sulcus bleeding index (P< 0.01 ).Furthermore, the sites with probing depth ≥4mm,attachment level ≥4mm were also more likely to detect Aa (P< 0.05 ,P< 0.01 ).Conclusion DNA probe assay is better than the traditional bacterial identification.If possible,the detection of Aa and suitable systemic antibiotics should be considered in adult periodontitis.
