摘要
目的比较3种不同门冬氨酸钾镁注射剂补充细胞内钾(K+)、镁(Mg2+)的效率及其对抗内毒素(LPS)肝细胞损伤的差异。方法常规培养肝细胞L02株,分为空白对照组、A组、B组和C组。除空白对照组外,各组细胞培养液中分别加入A、B、C三种门冬氨酸钾镁注射剂(均含K+5.3 mmol.L-1及Mg2+2.8 mmol.L-1),继续培养细胞1、2、3、5 h后,测定细胞内K+和Mg2+的含量。对抗内毒素(LPS)损伤实验中将肝细胞L02株分为LPS组、LPS+A组、LPS+B组、LPS+C组,各组均经终浓度为0.1 mg.L-1LPS和20 mmol.L-1的NH4Cl处理,同时将3种门冬氨酸钾镁注射剂分别加入各组,培养2 h后测定细胞培养液中尿素氮浓度和细胞膜Na+-K+-ATP酶的活性。结果与空白对照组比较,各门冬氨酸钾镁注射剂组K+、Mg2+进入细胞内的量均增多,2 h时A组的K+、Mg2+浓度明显高于B组和C组(P<0.05)。在对抗LPS损伤实验中,与LPS组比较,各门冬氨酸钾镁注射剂均可促进氨的清除,表现为尿素生成明显增多,以LPS+A组的效果最为显著(P<0.05);但3种门冬氨酸钾镁对内毒素损伤肝细胞膜Na+-K+-ATP酶活性的效应均不显著(P>0.05)。结论 3种门冬氨酸钾镁注射剂的生物学效应有一定差异,A组能较快地促进K+、Mg2+进入细胞,且显示出较好地拮抗LPS损伤肝细胞的作用。
AIM To investigate the effects of three different potassium aspartate and magnesium aspartate injections on the concentration of potassium(K+) and magnesium(Mg2+) in L02 cells and the protections against LPS damage to L02 cells.METHODS Cultured L02 cells were divided into four groups: control group,group A,group B and group C.Three different injections(containing 5.3 mmol.L-1 K+ and 2.8 mmol.L-1 Mg2+)were incubated into group A,B and C respectively.At 1,2,3 and 5 h after treatment,the content of K+ and Mg2+ in L02 cells were measured by inductively coupled plasma atomic emission spectrometers.To observe the protections against LPS damage,L02 cells were divided into four groups: LPS group,LPS + group A,LPS +group B and LPS + group C.LPS(0.1 mg.L-1) and NH4+Cl(20 mmol.L-1) were contained in culture media of all groups.After LPS treatment,three different injections were also incubated into group A,B and C,respectively.At 2 h,the activity of Na +-K +-ATPase and the concentration of urea in culture solution were measured.RESULTS The content of K+ and Mg2+ of L02 cells in three groups with potassium aspartate and magnesium aspartate injections were increased compare with the control group.Concentrations of K+ and Mg2+ in L02 cells of Group A was significantly higher than group B and C at 2 h(P 0.05).Compared with the LPS group,the NH3 was cleaned quickly by three potassium aspartate and magnesium aspartate injections.LPS +group A was significantly different with the other groups(P 0.05).However,there were no difference of the activity of Na+-K+-ATPase in all groups(P 0.05).CONCLUSION Three potassium aspartate and magnesium aspartate injections do have some different actions.Group A could improve the concentration of K+ and Mg2+ in L02 cells quickly,and protect L02 cells against LPS damage.
