摘要
目的:探讨钙离子螯合剂BAPTA-AM和钙离子载体A23187对胃癌SGC7901细胞葡萄糖调节蛋白78(Glucose regulated protein,GRP78)表达水平的影响,分析GRP78表达改变对胃癌细胞侵袭转移能力的影响。方法:BAPTA-AM和A23187处理细胞,RT-PCR和Western-blot检测3组细胞GRP78在mRNA和蛋白水平的表达,体外侵袭实验和划痕实验检测3组细胞的侵袭转移能力。结果:与对照组相比,BAPTA-AM组GRP78在mRNA和蛋白水平表达量降低,A23187组GRP78表达量升高;且BAPTA-AM组侵袭和转移能力明显低于对照组,而A23187组则高于对照组。结论:BAPTA-AM和A23187可分别下调和上调胃癌细胞GRP78的表达水平,GRP78的表达水平可能与胃癌细胞的侵袭和转移有关。
Objective:To investigate the effect of BAPTA-AM and A23187 on GRP78 expression of gastric carcinoma SGC-7901 cells in vitro,and the effect of GRP78 on invasion and metastasis of these tumor cells.Methods:The SGC7901 cells were treated by BAPTA-AM and A23187.RT-PCR and Western blot were used to assess the expression of GRP78 at both mRNA and protein levels.Matrigel invasion assay and a scratch wound assay were used to assess the invasion and migration levels.Results:The expression of GRP78 at both mRNA and protein levels,the invasion and migration levels were decreased in the BAPTA-AM-treated cells and increased in A23187-treated cells as compared with those of control group.Conclusion:BAPTA-AM can down-regulate the expression of GRP78 in SGC7901 cells and A23187 can up-regulate it.The expression of GRP78 in SGC7901 cells may be associated with their migration and invasion levels.
