电针对脾气虚大鼠肝组织细胞凋亡的影响

Effect on spleen-qi deficiency rats treated by electroacupuncture on apoptosis cells in liver

目的:从组织氧化与抗氧化损伤方面,观察电针对脾气虚大鼠肝组织细胞凋亡的影响,探讨电针治疗脾虚证的机理。方法:采用饥饱失常加劳倦过度法复制脾气虚模型;采用HE染色法检测大鼠肝组织细胞形态的变化,比色法检测实验大鼠肝组织丙二醛(Malondial dehyde,MDA)含量和超氧化物歧化酶(Superoxide dismutase,SOD)的活性,TUENL法定量检测实验大鼠肝组织细胞凋亡的表达。结果:脾气虚模型组大鼠肝组织中SOD活力较正常对照组明显下降(P<0.01),MDA含量较正常对照组显著上升(P<0.01);同时,脾气虚模型组细胞凋亡指数显著高于正常对照组(P<0.01)。电针治疗能上调肝组织中SOD活力,下调MDA含量,降低组织细胞凋亡数。结论:电针治疗脾虚证的机理可能与抗过氧化损伤,抗组织细胞凋亡的作用有关。

Objective：To observe the intervention effect on expression of apportosis and related factors of apportosis in spleen-qi deficiency rats treated by electroacupuncture in tissue oxidized or anti-oxidized injury and to investigate the protective effect and mechanism on spleen-qi deficiency treated by electroacupuncture.Methods：The rats＇ syndrome models of deficiency of spleen-qi were made by the method of compound causes of disease according to the TCM ＇s theory of injury due to diet and overstrain.Appearance of rats were observed：tissue ultrastructural changes measured with methods of Hemtoxycin-eosin（HE）.The activites of liver superoxide dismutase（SOD） and the contents of liver malondial dehyde（MDA） were determined by colorimetry.The apoptosis cells in liver tissue were tested by TdT—mediated digoxin—dUTP nick end labeling（TUNEL）.Results：We discuss the relationships between normal and model group,on the activity of SOD and the contents of MDA in liver of spleen-qi deficiency rats.The resu1t was that MDA of model group was more than normal group（P0.01） and the activity of SOD stronger of normal group（P0.01）.The apoptosis index was increased obviously in rat with liver injury induced by spleen-qi deficiency（P0.01）.The resu1t was decreased that the contents of MDA and the apoptosis index in live treated by electroacupuncture.The activity of SOD was stronger of electroacupuncture group.Conclusion：The Mechanism of electroacupuncture to spleen-qi deficiency maybe is regarded to the effect of Anti-Oxidanted injury and Anti-apoptesis in tissue cells.
