摘要
目的:观察鼻腔给予丙酸睾丸酮(TP)后大鼠行为以及黑质纹状体酪氨酸羟化酶(TH)和多巴胺转运体(DAT)表达的改变。方法:对正常成年雄性及去势大鼠鼻腔给予TP21d,分别利用开放广场实验和免疫组织化学技术观察给药后大鼠的行为及黑质和尾壳核TH及DAT的免疫反应强度变化。结果:开放广场实验结果显示正常大鼠鼻腔给予TP后水平运动得分、垂直运动得分、运动总径长比正常对照组增加;去势大鼠鼻腔给予TP后上述行为比去势组增加,但仍低于正常对照组。免疫组织化学结果显示正常大鼠鼻腔给予TP后黑质和尾壳核的TH及DAT免疫反应强度高于正常对照组;去势大鼠鼻腔给予TP后黑质和尾壳核的TH及DAT免疫反应强度高于去势组,但未达到正常对照组。结论:鼻腔给予TP增加大鼠的运动行为以及黑质和尾壳核TH和DAT的表达。
Objective: To observe the influence of intranasal administration with testosterone propionate (TP) on rat behaviors, tyrosine hydroxylase (TH) and dopamine transporter (DAT) in nigrostriatal system. Methods: Adult male wistar rats and castrated rats were treated by intranasal administration with TP for 21 days. Open field test and immunohistochemistry were respectively used to detect the altered behaviors and the altered immunoreactive intensity of TH and DAT in substantia nigra and caudate putamen of TP-treated rats. Results: The amount of locomotion activity (horizontal locomotion, vertical activity and total path length) was higher in adult male rats treated by intranasal administration with TP than that in the normal rats. While in the castrated rats treated by intranasal administration with TP, the amount of locomotion activity above was higher than that in the castrated rats, but still lower than that in the normal rats. Intranasal administration with TP to adult male rats was found to increase TH and DAT immunoreactive intensity in substantia nigra and caudate-putamen com- pared to normal male rats. The immunoreactive intensity of TH and DAT in substantia nigra and caudate putamen was stronger in castrated rats with intranasal TP than that in castrated rats, but weaker than that in normal male rats. Conclusion: Intranasal administration with testosterone propionate increased rat locomotion activity and the expression of TH and DAT in substantia nigra and caudate putamen.
