摘要
目的:观察急性脊髓损伤后中药髓复康干预对脊髓组织勿动蛋白-A表达的影响。方法:成年雄性恒河猴14只,其中10只用于制备T12胸髓左侧半横断损伤模型,并将其随机分为髓复康组和模型组,另外4只设为正常对照组。观察评估恒河猴脊髓神经功能恢复情况,采用原位杂交法及免疫组化法检测各组Nogo-A的表达量。结果:给药30d后,髓复康组患肢神经功能恢复优于模型组;脊髓损伤后Nogo-AmRNA及其蛋白的阳性细胞数及其阳性产物的OD值均明显升高,与正常组比较差异显著(P<0.05);其中髓复康组Nogo-AmRNA及其蛋白的阳性细胞数和其阳性产物的OD值都明显低于模型组。结论:中药髓复康对脊髓损伤后恒河猴脊髓中Nogo-A的表达有一定的抑制作用,为神经的修复和再生提供有利的微环境,促进了脊髓损伤后恒河猴运动神经功能的恢复。
Objective To investigate the effects of Chinese Herbal Medicine Suifukang (SFK)(髓复康) on the changes of the functional recovery and the expression of Nogo-A following spinal cord injury in rhesus monkeys. Methods There were fourteen adult rhesus monkeys and ten of them received left spinal cord hemisection at T12, and then randomly divided into two groups: SFK group (5) and control group (5). The other 4 monkeys remained in the normal group. The expression of Nogo-A mRNA was detected by in-situ hybridization and the expression of Nogo-A protein was detected by immunohistochemistry. Results The functional recovery of the nerve of the affected limb in the SFK group was better than that in the model group; The positive cells of Nogo-A mRNA and its OD value in the model group were more than in other groups (P 0.05); The Nogo-A mRNA positive cells and its OD value in SFK group were more than those in normal group (P 0.05). The changes of Nogo-A protein positive cells number and its OD value were similar to those of Nogo-A mRNA. Conclusion SFK can restrain the expression of Nogo-A obviously. It is possible to improve the microenvironment in the injured spinal cord for regeneration of nervous tissue, and facilitate the movement ability of injured spinal cord in rhesus monkeys.
