感染人巨细胞病毒后涎腺导管上皮细胞的病理形态学改变 被引量:1

摘要 目的:探讨涎腺导管上皮细胞感染人巨细胞病毒(HCMV)后的病理形态学改变。方法:应用免疫组化法标记11例腮腺巨细胞包涵体病尸检蜡块中HCMV感染细胞;应用HE染色、倒置相差显微镜和透射电镜从体内外观察HCMV感染细胞的形态学改变。结果:组织内HCMV感染细胞从未感染时的柱状变为圆形或椭圆形,体积增大,出现包涵体,近管腔处胞浆内见少量颗粒状物。体外培养的涎腺导管上皮细胞感染HCMV后,先是胞体皱缩变为球形,核略增大,以后细胞逐渐从培养瓶底部脱落。与体内感染相比,体外HCMV感染可导致宿主细胞在大小、形状、与周围细胞的连接和包涵体等方面出现差别。结论:体内外HCMV感染涎腺导管上皮细胞导致的形态学改变不同。 Objective:To study the pathological changes induced by human cytomegalovirus(HCMV)in duct epithelial cells of salivary gland.Methods:By immunohistochemical method,HCMV-infected cells were localized in tissues of 11 cases of parotid cytomegalic inclusion disease which were got by autopsy and embeded in paraffin.Morphological changes of HCMV-infected cells were observed in vivo and in vitro through HE staining,inverted phase contrast microscope and transmission electron microscope.Results:The shape of HCMV-infected cells changed from original stylolitic to round or elliptic.The enlargement of volume,inclusion and a small quantity of stippling at the lumina edge of cytoplasma were also found.After infected by HCMV,human salivary gland duct cells(HSG)cultivated in vitro were crimpled to be globular at first with the size of nucleus increased slightly and then detached from the bottom of culture flask.Compared with infection in vivo,there existed some differences in the size,shape,connection with periphery and inclusion of the host cells infected by HCMV in vitro.Conclusion:Morphological changes induced by HCMV in duct epithelial cells of salivary gland in vivo and in vitro are different.
