摘要
目的评价PBS、SM液、营养肉汤3种不同的采样液对分枝杆菌噬菌体D29的收集能力;D29在3种不同喷雾介质(生理盐水、去离子水和PBS)中的耐雾化性能;PBS作为采样液时,收集到的D29浓度随时间变化趋势。方法用全玻璃液体冲击式采样器AGI-30收集D29气溶胶并进行判定。结果与结论噬菌体D29在含有微量Tween80的PBS中耐冲击性能最强,将其用于采样能够准确反映气溶胶的浓度,而去离子水作为喷雾介质时,D29气溶胶具有最大的产出量。在D29气溶胶的动物暴露实验中,选择去离子水为喷雾液用于产生气溶胶,采样液为含微量Tween80的PBS。
Objective To evaluate the efficency of three sample liquid(PBS with Tween80,SM buffer,nutrient broth)in D29 aerosol sampling experiments.To select the best spray liquid which can protect D29 from injury during nebulization,to dilute the bacteriophage stock by three media:deionized water,PBS and isotonic saline water,respectively,and to generate aerosols with the resultant suspensions.Methods The all-glass sampler AGI-30 was used to collect aerosols.The results were reflected by the number of phages in the sampler.Results and Conclusion The order of the amount of phage D29 collected by AGI-30 was PBS (with Tween80) SM buffer nutrient broth.The maximum viable D29 was collected when the spray liquid was deionized water,followed by PBS and isotonic saline solution.In future animal exposure experiments,the spray liquid and sample media will be deionized water and PBS (with Tween80),respectively.
