摘要
目的研究脓毒血症及增龄大鼠膈肌葡萄糖转运蛋白-1(glucose transporter-1,Glut-1)和葡萄糖转运蛋白-4(glucose transporter-4,Glut-4)mRNA的变化,进一步探讨脓毒血症时呼吸肌代谢及功能变化的机制。方法建立大鼠脓毒血症模型,应用RT-PCR方法检测大鼠膈肌Glut-1和Glut-4 mRNA表达变化。结果大鼠膈肌Glut-1 mRNA和Glut-4 mRNA有增龄升高的变化趋势。大鼠膈肌Glut-1 mRNA和Glut-4 mRNA有随罹患脓毒血症升高的变化趋势。结论脓毒血症时膈肌的糖代谢失衡是导致呼吸功能障碍的原因之一,尤其是Glut-1和Glut-4的异常表达发挥作用,而且年龄因素也是影响Gluts表达的因素。大鼠膈肌Glut-1、Glut-4 mRNA表达具有特殊的增龄趋势,糖代谢失衡在老年脓毒血症大鼠中表现尤为明显,因此积极纠正糖代谢失衡可增加脓毒血症呼吸衰竭救治成功率。
Objective To investigate the expression of Glut-1 and Glut-4 mRNA in septic rat diaphragmatic muscle.Methods A rat model of septic and control model was established,and the expression of Glut-1 and Glut-4 mRNA in diaphragmatic muscle of SD rats were analyzed by RT-PCR.Results The expression of Glut-1 and Glut-4 mRNA showed an increasing trend with aging and sepsis.Conclusion The imbalance of diaphragmatic carbohydrate metabolism in sepsis plays a vitally important role in sepsis in respiratory dysfunction,especially,the Glut-1 and Glut-4 mRNA expressions in rat diaphragm are altered with age while sepsis effects on Glut-1 and Glut-4 mRNA expressions are age dependent.
