三苯氧胺对人子宫内膜癌细胞系Ishikawa细胞增殖的影响

摘要 目的:探讨三苯氧胺(TAM)在体外对Ishikawa人子宫内膜癌细胞株增殖的影响及其作用机制。方法:采用MTT法观察不同浓度TAM作用不同时间对Ishikawa细胞增殖的影响,另应用MTT法、流式细胞术、免疫细胞化学方法观察比较TAM、17β-雌二醇(17β-E2)及两者联合作用不同时间后对Ishikawa细胞增殖率,细胞周期时相分布以及C-myc、bcl-2、Bax 3种蛋白表达的影响。结果:TAM对Ishikawa细胞的促增殖作用在一定剂量范围内有浓度依赖性,可使G0/G1期细胞比例下降,S期细胞比例升高;C-myc、bcl-2蛋白表达增加,Bax蛋白表达下降。结论:体外TAM可能通过调节细胞周期时相分布及上调C-myc蛋白、bcl-2蛋白,下调Bax蛋白表达,促进Ishikawa人子宫内膜癌细胞增殖。 Objective:To explore the effect of tamoxifen(TAM)on proliferation of human endometrial carcinoma cell line Ishikawa cells and its mechanism.Methods:Ishikawa cells were treated with TAM,17β-estradiol(17β-E2)and TAM+17β-E2 respectively.MTT assay was used to observe the proliferation of Ishikawa cells after TAM treatment at different time with different concentrations;flow cytometry was adopted to observe the cell cycle phase distribution and the expression of C-myc,bcl-2,Bax were determined by immunocytochemistry method.Results:At some concentration range(10-8~10-6mol/L),TAM promoted the proliferation of Ishikawa cells in a dose-dependent manner.TAM blocked G0/G1 phase in cell cycle while stimulated S phase.TAM increased the expression of C-myc and bcl-2,while decreased the expression of Bax.Conclusions:TAM promoted the proliferation of Ishikawa cells in vitro by regulating the cell proportion at G1/S phases,up-regulating the expression of C-myc,bcl-2 and down-regulating the expression of Bax.
