戊己丸提取物不同配伍的肠外翻吸收研究 被引量:15

摘要 目的研究戊己丸不同配伍比例时在肠道中的吸收动力学的变化,说明方剂配伍的规律性。方法利用L9(34)正交设计,将黄连、制吴茱萸、土炒白芍组成不同配伍的9个组方,通过体外肠外翻模型和LC-MS分析手段,观察方中代表成分小檗碱、巴马汀、吴茱萸碱、吴茱萸次碱、芍药苷的吸收变化情况。结果以各代表成分的Ka为指标,通过正交分析,得出同一成分在肠道不同部位的吸收趋势大致相同,但不同成分之间的吸收趋势变化较大。多指标归一加权综合评分法分析表明,黄连在各肠段吸收贡献度最大,确定了其君药地位,制吴茱萸和土炒白芍次之,为臣药。在各个肠段的最优配比组合(黄连∶制吴茱萸∶土炒白芍)为:空肠12∶6∶12,回肠12∶6∶6与12∶6∶12,结肠12∶6∶12。结论戊己丸各代表成分在肠道中吸收动力学过程,随方剂的配伍比例不同而变化,其在肠道吸收的最优配伍组合为黄连∶制吴茱萸∶土炒白芍=6∶3∶6。 Aim To research the intestinal absorption at the different intestines using the different compatibility of extractive Wujiwan,in order to indicate the regularity of prescription compatibility.Methods The experiment was carried out with the L9(3 4 ) orthogonal design,which took the extracts of Rhizoma Coptidis,Evodiae Fructus and Radix Paeoniae Alba into nine different combinations.Each compatibility was studied by means of everted gut sacs.Five representative compositions of extractive Wujiwan berberine,palmatine,evodiamine,rutaecarpine and paeoniflorin were detected by LC-MS,and their changes of intestinal absorption were observed.Results The statistics was taken Ka as the marker and carried out by orthogonal analysis.The results showed that the same compound had the similar absorption trend at different intestines,and the different compounds were obviously various in absorption. Weight Grade method of normalization multicriteria indicated the extract of Rhizoma Coptidis made the greatest contribution at different intestines which determined it monarch,and the extract of Evodiae Fructus and Radix Paeoniae Alba were minister.The optimized compatibility at different intestines was 12∶6∶12 at jejunum,12∶6∶6 and 12∶6∶12 at ileum,12∶6 ∶12 at colon,respectively.Conclusions The absorptive kinetics of the representative compositions in Wujiwan expressed diversity in different compatibility at different intestines,and the best optimized absorption compatibility was 6∶3∶6(extractive Rhizoma Coptidis∶extractive Evodiae Fructus∶extractive Radix Paeoniae Alba).
