老年大鼠局灶脑缺血半影区小胶质细胞可塑性变化和bFGF表达规律的实验研究 被引量:2

摘要 目的观察青年大鼠和老年大鼠局灶性脑缺血半影区小胶质细胞形态学变化及bFGF的表达规律。方法老年(20~22月龄)雄性SD大鼠36只,青年(4~5月龄)雄性SD大鼠36只;假手术组12只(老年、青年大鼠各6只),脑缺血组60只(老年大鼠和青年大鼠各30只),采用大鼠大脑中动脉闭塞(middlecerebral artery occlusion,MCAO)模型,根据缺血的时间不同分为缺血后3、7、14、21、32d5个亚组,毎亚组各6只;分别取脑,行HE染色和免疫组化染色,观察小胶质细胞的变化和BFGF的表达水平。结果在局灶脑缺血的不同时期老年大鼠半影区小胶质细胞和bFGF的表达水平均低于青年大鼠,且表达高峰延迟。结论与青年大鼠比较,老年大鼠脑缺血后局部损伤严重,修复缓慢。 Objective To observe the morphology change of microglia and the expression of bFGF in cerebral ischemia penumbra of young and old rats after permanent middle cerebral artery occlusion.Methods Thirty-six young male Wistar rats and thirty-six old Wistar rats were tested.Twelve of them(young 6 and old 6)for false operation group,sixty of them(young 30 and old 30)for permanent focal cerebral ischemia group.The latter were divided into five subsections(ischemia for 3 days,7 days,14 days,28 days and 42 days) according to ischemia time.HE staining and immunohistochemical staining were underwent respectively.The morphology change of microgli and the expression of bFGF were observed.Results Microglia and the expression of BFGF in different periods of old rats were lower than that of young rats,and the peak of expression delayed.Conclusions The more serious injure and the slower repair in old rats than in youngrals after permanent focal cerebral ischemia.
