灵芝原生质体单核化杂交子的评价 被引量:11

Assessment of hybrids obtained from Ganoderma lucidum by using protoplast monokaryon technique

摘要 本研究对比分析了3个灵芝原生质体单核化杂交子及其亲本的菌丝生长速度、产量、出菇期、有效成分含量;评价了采用原生质体单核化技术进行灵芝杂交育种的可行性.测定结果表明,3个杂交菌株中,"杂交1号"分别比亲本"广东赤芝"和"灵芝5.75"提早22和3 d出菇,产量比"广东赤芝"高178.5%,比"灵芝5.75"高34.9%;"杂交2号"的菌丝生长速度明显快于2个亲本菌株,子实体三萜含量比"广东赤芝"高198%,比"灵芝5.75"高121.2%;这2个杂交子表现出明显的杂交优势,说明这种灵芝原生质体单核化杂交育种技术有实用价值,可用于灵芝新品种培育. The monokaryon strains were isolated by using protoplast monokaryon technique,and then cross paired to obtain hybrids.The mycelial growth rate,fruiting time,yield and activity ingredient of 3 hybrids and their parents were compared and anlyzed.The results showed that the fruit bodies of Hybrid-1 appeared 22 days and 3 days earlier than their parent of "Guangdong Chizhi" and "Ganoderma 5.75" respectively,and the yield of Hybrid-1 was 178.5% higher than "Guangdong Chizhi",34.9% higher than "Ganoderma 5.75".Compared with two parent strains,Hybrid-2 had faster growth rate at 30 ℃ in PDA medium.The triterpenoids of Hybrid-2 was 198% higher than "Guangdong Chizhi" and 121.2% higher than "Ganoderma 5.75" respectively.The vigor of the hybrids was determined in our experiment.The results indicated that this technique was valuable and could be applied in the hybreeding of Ganoderma lucidum.
