肿瘤坏死因子相关凋亡诱导配体联合相关小分子药物对胰腺癌细胞的抑制作用

Inhibition effects of TRAIL combined with small molecule drugs on pancreatic cancer cells

摘要 目的探讨肿瘤坏死因子相关的凋亡诱导配体(TRAIL)联合相关小分子药物对胰腺癌细胞的抑制作用。方法运用MTT、流式细胞仪和Western blot方法观察TRAIL、白藜芦醇、紫杉醇和三氧化二砷(或药物组合)对人不同胰腺癌细胞系的敏感性及协同效应。结果单独使用TRAIL、白藜芦醇或紫杉醇没有引起capan-2细胞明显的凋亡。将TRAIL和白藜芦醇或紫杉醇联合使用,均引起capan-2细胞的明显凋亡;其对colo-357细胞也有很强的协同作用。几种药物对Miapaca-2细胞都有较明显抑制作用;但在capan-1细胞中,几种药物基本无协同作用。结论 TRAIL与白藜芦醇或紫杉醇联用可以提高TRAIL对胰腺癌细胞的凋亡诱导效果。 Objective To explore the inhibition effects of TRAIL combined with small molecule drugs on pancreatic cancer cells.Methods The effects of TRAIL or combined with paclitaxel or resveratrol on pancreatic cancer cellines were detected by MTT assay,flow cytometry and Western blotting.Results Apoptosis of Capan-2 cell was not significantly induced by TRAIL,resveratrol or taxol alone.Combined use of TRAIL with resveratrol or paclitaxel produced significant TRAIL-induced apoptosis of Capan-2 celline and colo-357 cell.Combined use of TRAIL with resveratrol or paclitaxel had a synergistic inhibitory effect on Miapaca-2 cell,but not on capan-1 cell.ConclusionTRAIL combined with resveratrol or paclitaxel can enhance TRAIL induced apoptosis in pancreatic cancer
