摘要
目的用自建的流式微球分析技术(CBA)对急性心肌梗死(AMI)患者和健康对照组进行肌钙蛋白T(cTnT)检测,并与电化学发光法进行比较,观察其临床应用价值。方法用鼠抗人cTnT的单克隆抗体包被在已激活的羧基化聚苯乙烯微球,使用羊抗人cTnT的多克隆抗体与cTnT结合,标记在驴抗羊多克隆抗体上的异硫氰酸荧光素(FITC)为指示剂,建立CBA。同时以电化学发光法作对照。结果 AMI组和对照组血清cTnT经CBA方法和电化学发光法检测,2组结果差异均有统计学意义。CBA方法和电化学发光检测结果比较差异无统计学意义,2种方法的回归方程为Y=0.987X+0.013,r=0.9976,具有显著的相关性。结论自建CBA检测技术检测cTnT,与电化学技术检测结果比较差异无统计学意义,具有显著的相关性,可在临床工作中推广使用。
Objective To detect the troponin T(cTnT)of acute myocardial infarction(AMI)patients and healthy controls by self-built cytometric bead assay(CBA),and compared with the result of electroche-miluminescence.Methods The CBA was established with that the already activated carboxylated polystyrene microspheres was coated with mouse anti-human cTnT monoclonal antibody,the goat anti-human cTnT polyclonal antibody was used to combine with the cTnT as the second antibody,and the fluorescein isothiocyanate(FITC) labeled donkey anti-goat polyclonal antibody as the indicator.The results of electrochemiluminescence were as control.Results The serum content of cTnT in AMI group and control group detected with CBA and electrochemiluminescence were significantly different.There were no significant differences between the detection results of the CBA and electrochemiluminescence,and the regression equation of the two methods were:Y=0.987X+0.013,r=0.997 6,the correlation was significant.Conclusion Self-built CBA had no difference with electrochemiluminescence in detecting cTnT,and the correlation was significant,so it could be used widely in clinical practice.
