高温致神经管畸形金黄地鼠HSP70及p38MAPK的通路变化

HSP70 and p38MAPK Pathway Change in Neural Tube Defects Induced by Hyperthermia in Golden Hamster

摘要 目的探讨热休克蛋白70(HSP70)及p38MAPK信号通路蛋白在高温致神经管畸形中的机制。方法将清洁级雌性金黄地鼠孕鼠于妊娠第8天进行水浴,分别于高温后的8、16、24、48、72、96h取出36只正常对照组孕鼠(水浴温度为37℃,持续20min)和36只高温致神经管畸形的实验组孕鼠(水浴温度为42℃,持续20min),取胚胎脑组织。应用Western blot检测HSP70和p38MAPK及P-p38MAPK蛋白的表达水平,RT-PCR法检测HSP70和p38MAPK的mRNA表达。结果高温处理后HSP70mRNA及蛋白表达水平在24~96h均不同程度地高于对照组(P<0.05);高温后8~48hp38MAPK磷酸化蛋白产物增加,于16h表达增加最为显著(P<0.01),而8~96h,对照组和实验组间p38MAPKmRNA及蛋白含量差异无统计学意义。结论 HSP70及p38MAPK信号通路蛋白的变化与高温致金黄地鼠神经管畸形密切相关。 Objective To study the role of HSP70 and p38MAPK pathway in the development of neural tube defects caused by hyperthermia.Methods The pregnant golden hamsters were treated with high temperature water bath at gestational day 8,for 8,16,24,48,72 and 96 hours respectively,and the animals were sacrificed,the embryonic brains in both of exposure and control groups were taken for detecting the protein expression of HSP70,p38 MAPK and P-p38 MAPK.The gene transcription of HSP70 and p38MAPK was measured by RT-PCR.Results Compared with the control,in the hyperthermia group,the expression of mRNA and protein of HSP70 increased at 24-96 h(P〈0.05);p-p38MAPK protein increased at 8-48 h,and the peak appeared at 16 h(P〈0.05).No significant change was observed in the gene and protein level of p38MAPK and protein.Conclusion The change of HSP70 and p38MAPK pathway may be strongly related to hyperthermia induced neural tube defects in golden hamster.
