摘要
建立了反相高效液相色谱法分离和测定维生素C的方法。使用C8色谱柱(150mm×4.6mmi.d.,5μm)作为固定相,通过对流动相的缓冲盐种类、pH值、有机溶剂的比例等实验条件的考察,确立了以pH2.5的磷酸缓冲液/甲醇(98/2,v/v)作为流动相,246nm作为检测波长的最佳色谱条件。用该检测方法对维生素C与苯甲酸钠的共存情况进行考察,发现在不同pH值情况下,维生素C与苯甲酸钠的水溶液在80℃共存2h,两者不发生反应。
A reversed-phase performance liquid chromatographic method was developed for theseparation and determination of vitamin C.Zorbax C8 column(150 mm × 4.6 mm id,5 μm) was used asstationary phase.By examining the types of buffer salt,the different pH values and different proportion oforganic solvent,pH2.5 phosphate buffer:methanol /98:2(v/v) mixture was selected as mobile phase.246nm was adopted as the best chromatographic condition for wavelength detection.With this detectionmethod,the coexistence of Vitamin C and sodium benzoate was observed.It's found that at different pHvalues,vitamin C and sodium benzoate could coexist for 2 hours at 80 ℃ without reaction in aqueoussolution.
