摘要
采用一次性被动回避式反应方法跳台法和避暗法,以及水迷宫法和穿梭箱法,研究了蜕皮甾酮(ECR)对地西泮和乙醇所造成的记忆障碍的保护作用.结果表明,ECR1.8-14.4mg·kg-1能以倒U型量效曲线方式预防地西泮致小鼠健忘和乙醇造成的小鼠记忆再现缺失,ECR3.6,7.2mg·kg-1能拮抗乙醇引起的小鼠空间辨别障碍,ECR5.0mg·kg-1还能减轻乙醇引起的大鼠主动回避反应障碍.提示ECR能保护地西泮与乙醇造成的记忆损害。
The protective effect of ecdysterone (ECR) on amnesia induced by diazepam and alcohol was studied by using the method of the one once trial of passive avoidance response including step down test and step through test, water maze test and shuttle box test. The results showed that ECR 1.8-14.4 mg·kg 1 prevented the diazepam (1.25 mg·kg 1 , ig) induced amnesia and the retrieval deficit induced by alcohol (2.5 g·kg 1 , ig) in mice in an inverted U dose response curve manner, ECR 3.6 and 7.2 mg·kg 1 antagonized the spatial discriminate damage induced by alcohol (0.5 g·kg 1 , ip) in mice, ECR 5.0 mg·kg 1 also protected rats against the impairment of multiple training active avoidance response induced by alcohol (2.0 g·kg 1 , ip). However, in the effective dose range influencing memory, ECR had no effect on the spontaneous locomotor activity, the pentobarbital sodium induced sleeping at the sub threshold dose of 30 mg·kg 1 and painthreshold in mice. These results indicate that ECR has protective effect against amnesia induced by diazepam and alcohol, and the influence of ECR on memory may be related to its antagonistic actions against the positive modulation of diazepam or alcohol on the GABAergic nervous system.
