环孢霉素A对培养的人角朊细胞主要组织相容性抗原表达的影响

摘要 目的:研究环孢霉素A对人皮肤角朊细胞的主要组织相容性抗原(MHC)表达的影响.方法:采用γ-干扰素(INF-γ)刺激培养的人角朊细胞MHC-Ⅰ/Ⅱ抗原表达的实验模型,研究了低剂量环孢霉素A对角朊细胞MHC-Ⅰ/Ⅱ抗原表达的影响.结果:0.125mg/L的环孢霉素A对于INF-γ刺激的角朊细胞MHC-Ⅰ类抗原(HLA-ABC抗原)表达的增强,具有抑制作用(P<0.01);但是对于INF-γ刺激的角朊细胞MHC-Ⅱ类抗原(HLA-Dr,HLA-Dp抗原)表达的增强,没有抑制作用. AIM: To study the effect of cyclosporin A on the major histocompatibility complex(MHC) antigen expression of human skin keratinocytes. METHODS: The influence of low dose cyclsporin A on keratinocyte MHC Ⅰ/Ⅱ antigen expression was assessed with an experimental model in which the expression of cultured human keratinocyte MHC Ⅰ/Ⅱ antigen was induced by INF γ. RESULTS: 0.125 mg/L cyclsporin A inhibited INF γ induced expression of keratinocyte MHC Ⅰ antigen (HLA ABC antigen) whereas no inhibitory effect on INF γ induced expression of keratinocyte MHC Ⅱ antigen (HLA Dr, HLA Dp antigen) was observed. CONCLUSION: The mechanism underlying prolonged human allo graft survival by cyclsporin A is related to the inhibitory effect of cyclsporin A on the keratinocyte MHC I expression of donor skin.
