深Ⅱ°烧伤创面愈合过程中EGF/EGFR的表达 被引量:18

摘要 目的:了解人深Ⅱ°烧/烫伤创面愈合过程中创面内源性EGF及其受体的动态表达,以便为临床合理应用外源性EGF提供依据.方法:分别于伤后1,3,5,7,10,14及21d自深Ⅱ°烧/烫伤住院患者创面取材,同时取正常皮肤做对照.应用免疫细胞化学和原位杂交技术检测EGF/EGFR及其mRNA的表达.结果:EGF于伤后1d~5d较对照明显降低,伤后7d恢复至对照水平,伤后10d显著增加,并于伤后14d达到高峰值,伤后21d与对照无明显差别.EGFR于伤后1d~3d明显低于对照,伤后5d开始增加,并于伤后10d达到最大值,尔后仍以显著高于对照的量持续至伤后21d.EGFmRNA与EGFRmRNA分别于伤后10d和伤后7d~10d呈强阳性表达,其前后表达则较弱.结论:伤后早期或后期于创面局部应用外源性EGF也许有助于更有效的促进深Ⅱ°烧/烫伤创面愈合. AIM: To understand the dynamic expression of endogenous EGF and its receptor in deep second degree burn or scald wound in patients during wound healing in order to provide theoretical basis for clinical application of exgenous EGF. METHODS: The samples were harvested from second deep degree burn or scald wounds in patients on the 1st, 3rd, 5th, 7th, 10th, 14th and 21st day post injury respectively. At the same time, normal skin was harvested to be used as control. Immunohistochemistry and hybridization were employed to detect the expression of EGF and EGFR, and their mRNA in wounds as well. RESULTS: The expressed EGF showed a marked decrease compared with the control from the 1st day to the 5th day post injury. On the 7th day, the expressed EGF reached the same level as that of control. From the 10th day,it increased significantly and reached the peak value on the 14th day. It dropped to that of the control on the 21st day post injury. The expressed EGFR was obviously lower than that of the control in the first three days post injury and began to increase on the 5th day. The peak value was reached on the 10th day. On the 21st day, it was still markedly higher than that of the control. The expressed intensities of EGF mRNA and EGFR mRNA reached highest level on the 10th day and in period of 7 days to 10 days post injury respectively. CONCLUSION: The application of exogenous EGF on wounds in the early and later stages in second deep degree burn or scald may accelerate wound healing.
