摘要
目的:建立测定氯碘羟喹乳膏含量的HPLC法。方法:采用C18固相萃取小柱分离除去氯碘羟喹乳膏中的基质,高效液相色谱法测定氯碘羟喹的含量。色谱柱为Zorbax C8柱(4.6mm×150mm,5μm);流动相为乙二胺四醋酸二钠盐缓冲液(取乙二胺四醋酸二钠盐0.5g,加水400mL,振摇使溶解,加三乙胺4mL,用磷酸调节pH值至3.0)-甲醇(35∶65),检测波长为254nm,流速1.0mL·min-1。结果:氯碘羟喹进样量在3~60μg范围内呈良好的线性关系(r=0.99995),平均回收率为99.0%(n=9)。结论:与现行方法比较,该方法专属性强,适用于测定氯碘羟喹乳膏含量。
Objective:To establish a method for the determination of Clioquinol in Clioquinol Cream by HPLC. Methods: The sample was purified with C18 solid phase column and its content was determined with HPLC method. The Zorbax Cs column (4. 6 mm × 150 mm, 5 μm) was used and mobile phase was EDTA-2Na buffer solution (dissolving 0. 50g of EDTA-2Na in 400 mL of water, adding 4. 0 mL of triethylamine and mixing, using phosphoric acid to adjust pH value of 3. 0) - methanol (35:65). The detection wavelength was at 254nm, and the flow rate was 1.0 mL· min^-1. Results: Within the range of 3 -60 μg, peak area and clioquinol injection quantity were in linear Correlation (r =0. 999 95) and the average recovery were 99. 0% (n =9). Conclusion. In comparison to the previous methods, the new method was more specific and can control quality of product effectively.
