豆蔻提取物对环氧合酶-2在人胃癌裸鼠移植瘤中表达的研究 被引量:1

Effects of Fructus Amomi Rotundus Extraction on Cyclooxygenase-2 Expression of Nude Mice Model of Human Gastric Cancer Transplantation

摘要 目的观察豆蔻提取物对人胃癌细胞裸鼠移植瘤的抑制作用和对环氧合酶-2(Cycloozygenase-2,COX-2)表达的影响。方法建立人胃癌SGC-7901细胞裸鼠皮下移植瘤模型,随机分为对照组、豆蔻组、5-FU组、联合组,各组按设计剂量给药。测定移植瘤体积、质量及抑瘤率;用免疫组化法检测各组移植瘤瘤体中COX-2的表达情况。结果豆蔻组、5-FU组、联合组的平均瘤质量和瘤体积明显低于对照组(P<0.05),3组抑瘤率分别为45.83%、63.67%、79.25%。联合组瘤体中COX-2表达的程度明显低于对照组(P<0.05)。结论豆蔻提取物可抑制胃癌移植瘤的生长,与5-FU联合应用具有协同作用,其作用机制可能与下调瘤灶中COX-2的表达有关。 Objective To observe the effects of fructus amoral rotundus extraction on the growth of transplanted tumor and expression of cyclooxygenase-2 (COX-2) on nude mice model of transplantation of human gastric cancer. Methods A mouse tumor model was set up by implanting SGC-7901 human gastric cancer cells under the skins of nude mice. Twenty-four experimental mice were randomly divided into four groups (six in each): control group, 5-fluorouracil group (5-FU group), fructus amomi rotundus extraction group, combination group. For each group, the drug was administered with designed dose. The volume and weight of transplantation tumor were measured, and the tumor inhibition rate was calculated. The expression of COX-2 in tumor was detected in immunohistochemisty way. Results The weight and volume of the tumors in 5-FU group, fructus amomi rotundus extraction group and combination group was less than that of the control group significantly, The tumor inhibition rate of the three groups was 45.83%, 63.67%, 79.25% respectively (P〈0.05). The expression of COX-2 in combination group was less than that of the control group. Conclusion Fructus amomi rotundus extraction can inhibit the growth of tumor and markedly enhance the inhibitory effect by combining with 5-FU. The possible mechanism may be related with down-regulating the expression of COX-2.
