摘要
单因素试验分析和正交试验确定微波辅助提取连翘花黄色素的工艺条件,Fenton法测定连翘花黄色素的抗氧化活性。结果显示,影响色素提取的因子依次为提取时间>提取剂浓度>微波功率>料液比,提取的最佳条件为B2D3A3C2,即使用95%vol乙醇,以料液比1∶40,微波功率700W,每次提取时间40s。以此工艺条件提取连翘花黄色素,产率为10.54%,黄色素的95%vol乙醇溶液在可见光区的最大吸收波长为410nm,该色素溶液还原力强,具有清除.OH的能力,在色素浓度为10.0mg/mL时,对.OH的清除率为97.67%。
Extraction technology of yellow pigment from Forsythia suspensa (Thunb.) vahl with microwave assisted method was determined with single-factor and orthogonal experimental design. Antioxidant activity of extracted pigment was evaluated with Fenton method. The results showed that the factors affected extraction rate were ordered as extraction time 〉 alcohol concentration of solvent 〉 power of microwave 〉ratios of raw material to solvent. The optimum conditions were 95% alcohol, ratios of raw material to solvent were 1:40, power of microwave 700W, extraction time 40s, and under these conditions, yield of yellow pigment was 10.54% and hmax was 410nm. The yellow pigment had reducing activity and capacity of scavenging of free radical (· OH). Rate of scavenging on · OH was 97.67% when the concentration at 10.0mg/ml.
