胃癌组织中核因子κB和环氧合酶2的表达及其与幽门螺杆菌感染的关系 被引量:6

Expression of NF-κB and COX-2 in Human Gastric Carcinomas and Their Relationship with H.pylori Infection

摘要 目的观察胃癌组织中核因子κB(NF-κB)和环氧合酶2(COX-2)的表达,探讨二者与幽门螺杆菌(H.pylori)感染的关系。方法采用免疫组织化学染色法(SP法)检测10例正常胃黏膜组织及50例胃癌患者手术切除胃黏膜组织中NF-κB和COX-2的表达,用快速尿素酶诊断法和13C呼气试验两种方法检测胃癌患者H.pylori感染状况,分析NF-κB和COX-2的表达及H.pylori感染与胃癌的关系。结果胃癌组织NF-κB和COX-2的阳性表达率〔分别为74%(37/50)和86%(43/50)〕显著高于正常胃黏膜组织(分别为0/10和1/10),差异有统计学意义(P<0.01)。胃癌组织中NF-κB与COX-2的表达呈正相关(r=0.524,P<0.01)。H.pylori感染者胃癌组织中NF-κB、COX-2的阳性表达率〔分别为93%(27/29)和86%(25/29)〕显著高于未感染者〔分别为52%(11/21)和48%(10/21)〕,差异有统计学意义(P<0.05)。结论胃癌组织中NF-κB和COX-2的表达增加,且NF-κB与COX-2表达呈正相关;H.pylori感染可能上调胃癌组织中NF-κB、COX-2的表达。 Objective To detect the of expression of nuclear factor-kappa B(NF-κB) and cyclooxygenase 2(COX-2) in the tissue of gastric cancer and discuss their relationship with Helicobacter pylori(H.pylori) infection.Methods The tissues from 50 cases of gastric carcinoma and 10 cases of normal tissues were evaluated to detect the expression of NF-κB and COX-2 by SP immunohistochemistry and H.pylori infection was detected by fast urea enzyme test and 13C breath test.Analyze the expressions of NF-κB and COX-2,and the relationshiop between H.pylori infection and gastric carcinoma.Results The positive rate of NF-κB and COX-2〔respectively 74%(37/50)and 86%(43/50)〕in gastric carcinoma was significantly higher than that in normal tissues(respectively 0/10 and 1/10)(P0.01).NF-κB expression was positive correlated with COX-2 expression in gastric carcinoma(r=0.524,P0.01).The positive rate of NF-κB and COX-2 in gastric carcinoma with H.pylori infection〔respectively 93%(27/29)and 86%(25/29)〕was significantly higher than that in gastric carcinoma without H.pylori infection〔respectively 52%(11/21) and 48%(10/21)〕(P0.05).Conclusion Expression of NF-κB and COX-2 is increased and NF-κB expression is positive correlated with COX-2 expression in gastric carcinoma;H.pylori infection may up-regulate the expression of NF-κB and COX-2.
