摘要
目的探讨姜黄素对人胃低分化腺癌MGC-803细胞株增殖及细胞中核因子κB(NF-κB)蛋白表达的影响。方法将培养的MGC-803细胞分为对照组(A组)、5μmol/L姜黄素作用组(B组)、10μmol/L姜黄素作用组(C组)和20μmol/L姜黄素作用组(D组),分别干预24、48、72h,用cellcounting Kit-8(CCK-8)法检测细胞增殖抑制情况,用免疫蛋白印迹(Western blot)法检测各组作用24h后细胞中NF-κB表达情况。结果作用24、48、72h,不同浓度的姜黄素对胃癌细胞MGC-803的增殖有抑制作用,且随姜黄素浓度的增加和作用时间的延长,增殖抑制率逐渐增加。Western blot法检测显示,作用24h后A、B、C、D组MGC-803中NF-κB蛋白表达量分别为(0.889±0.019)、(0.640±0.019)、(0.503±0.014)和(0.349±0.022),随着作用浓度的增加,NF-κB表达量逐渐降低,组间差异均有统计学意义(P<0.01)。结论姜黄素能够抑制胃癌细胞MGC-803增殖,且随着姜黄素浓度的增加和作用时间的延长,细胞增殖抑制率逐渐增加;胃癌细胞MGC-803存在NF-κB的表达;姜黄素通过下调NF-κB的表达从而对胃癌细胞MGC-803的增殖起到抑制作用。
Objective To study the expression of nuclear factor κB(NF-κB) in gastric cancer cells MGC-803 before and after the treatment with curcumin.Methods Cultured gastric cancer cells mgc-803 was divided into control group(group A),5 μmol/L curcumin group (group B),10 μmol/L curcumin group(group C) and 20 μmol/L curcumin group(group D),all of which were treated for 24h,48h,72h respectively.The inhibition of cell proliferation was evaluated by CCK-8(Cell Counting Kit-8) and western blot was used to examine the expression of NF-κB protein in each group after 24h.Results Curcumin could inhibit the proliferation of gastric cancer cells MGC-803 and the inhibition rate depended on time and the concentration of curcumin.Western blot analysis demonstrated that the expression level of NF-κB protein decreased with the increase of drug concentration,the difference between group A (0.889±0.019),group B (0.640±0.019),group C(0.503±0.014) and group D(0.349±0.022) was significant (P0.01).Conclusion Curcumin could inhibit the proliferation of gastric cancer cells MGC-803,which depended on the effect time and the concentration of curcumin.There were expression of NF-κB in gastric cancer cells.Curcumin inhibited the proliferation of gastric cancer cells MGC-803 through down-regulating the expression of NF-κB.
