摘要
目的研究烧伤情况下骨髓造血基质细胞支持粒单系造血的功能变化。方法采用体外小鼠骨髓基质细胞融合培养和粒单系祖细胞集落(CFU-GM)培养的方法,动态观察15%Ⅲ度烧伤时骨髓基质细胞对粒单系祖细胞生长的影响。结果当在烧伤组基质细胞层上培养正常骨髓细胞的CFU-GM时,其集落生成能力在伤后1,3,5,7,10天均明显低于正常对照组,而同时培养的烧伤小鼠骨髓细胞CFU-GM集落则为伤后1~5天显著降低,第7天与正常组无差别,第10天时明显超过正常,与正常基质细胞层上培养烧伤组CFU-GM时有相同的变化趋势。结论烧伤时骨髓基质细胞支持粒单系造血的功能受到了一定程度的损伤,且较难恢复,而粒单系祖细胞的损伤则恢复较快。
Objective
To investigate the effect of bone marrow stromal cells of mice with 15% TBSA degree burn on
the growth of granulo-monopoietic progenitors. Methods The culture and confluent of the stromal
cell of the mice bone marrow and semi-solid culture of the colony-forming unit of
granulo-monopoietic progenitors (CFU-GM) in vitro were employed. Results The number of
normal bone marrow CFU-GM on the surface of stromal cells layer in burned mice was
significantly reduced as compared with controls at the days 1,3,5,7,10 post injury;whereas the
burned mice bone marrow CFU-GM under the same cultural condition were also decreased at
the days 1-5,but at days 7 and 10 the number was recovered and increased,respectively.The
differences of the effect of bone marrow stromal cells and GM-CSF on the growth of CFU-GM
were also investigated. Conclusions The supporting function of bone marrow stromal cells on
CFU-GM is impaired under the condition of 15% TBSA degree burn in mice.
