槐花总黄酮提取物HPLC指纹图谱研究

HPLC Fingerprint Spectrum of the Flavonoids from Sophora Japonica L.

目的:建立槐花总黄酮提取物的高效液相色谱指纹图谱,为科学评价及有效控制其质量提供可靠方法。方法:采用高效液相色谱法,以DIKMA C18(5μm,4.6mm×250mm)为色谱柱,甲醇-1%冰醋酸为流动相梯度洗脱,流速1.0mL.min-1,检测波长257nm,分析时间70min。测定10批样品色谱图,建立指纹图谱。结果:确定了17个共有峰,其中7号、12号峰分别为芦丁、槲皮素。10批样品相似度均大于0.90。结论:建立的HPLC指纹图谱分析方法可用于槐花总黄酮的质量评价。

Objective：To establish the fingerprint of total flavonoid from Sophora japonica L.by HPLC,provide a reliable method for the science appraisal and active control its quality.Methods： HPLC with DIKMA C18 chromato bar（5μm,4.6mm×250mm）,using gradient elution with methanol-1% glacial acetic acid,under the detection wavelength of 257nm,volume flow rate 1.0mL·min-1 and record time 70 min.Chromatogram of 10 samples determined to establish fingerprints.Results：17 common peaks in the fingerprints were positioned with index component of flavonoids,to identify rutin and quercetin.Average similarity of 10 samples was over 0.90.Conclusion： This HPLC method could be used to obtain the fingerprint of total flavonoid from Sophora japonica L.and control the quality.
