富含甘油三酯的脂蛋白中载脂蛋白B-48检测方法的建立

Development of Quantitation Method for Apolipoprotein B 48 in Triacylglycerol rich lipoproteins

用特异性的酶联免疫吸附法（ＥＬＩＳＡ）建立富含甘油三酯的脂蛋白中载脂蛋白Ｂ－４８（ａｐｏＢ－４８）含量的检测方法。研究表明：特异性的ａｐｏＢ－４８抗血清能够识别脂蛋白颗粒表面的蛋白质Ｃ末端。检测下限为３７μｇ／ｍｌ。变异系数板内为３．８％，板间为８．６％。回收率为（１０２±１０）％。ａｐｏＢ－４８抗血清与人体血清白蛋白、卵白蛋白、甲状腺球蛋白及载脂蛋白Ｂ－１００（ａｐｏＢ－１００，用免疫吸附层析纯化）没有交叉反应。本法特异性强，灵敏度高，简便快捷。

By using a specific antiserum for apolipoprotein B 48 in a competitive,enzymelinked immunosorbent assay(ELISA),we developed the quantitatiion method for apolipoprotein B 48 in triacylglycerol rich lipoproteins.The antiserum act as an effective immunoblotting agent and its use in this ELISA indicates that the antiserum recognizes the C terminal region of the protein on the surface of lipoprotein particles.Validation of the assay,included intra and inter assay reproducibility,sensitivity,parallelism,recovery of a known amount of apoB 48 form a spiked sample and crossreactivity,were performed on the ELISA.
