摘要 烤烟叶片还原糖含量是烤烟品质的重要指标之一,现有的测定方法在稳定性、准确度、成本消耗及效率方面尚存在不足之处.该研究在对3,5-二硝基水杨酸(DNS)法与斐林试剂比色法进行比较的基础上,对3,5-二硝基水杨酸(DNS)法提取烤烟叶片还原糖含量进行了优化.结果表明:可将预浸提试剂由酒精改为蒸馏水,浸提时间由10 h缩短为10 min;对于大量样品检测而言,此方法的改进可简化流程,节约成本,提高效率. Reducing sugar content is one of the important indicators of quality of flue-cured tobacco.There are some shortcomings in existing determination methods of reducing sugar content in flue-cured tobacco leaves.In this study,3,5-dinitrosalicylic Acid(DNS) Method for determining the reducing sugar content in the leaves of tobacco was improved and applied,based on comparing with the colorimetry method of Fehling reagent.In the improved DNS method,alcohol used to the pre-extracting was instead of distilled water and pre-extracting-time was shortened to 10 min.The improved method was proved to be much simpler,more economical and efficient in testing a large number of samples.
